Another less severe thing to help with the need to poop is Pepto (or generic equivalents). I keep a few chewable tablets in my hydration vest.
And other suggestions for a first 100 miler:
* Some plan can help crew if you have one — they need to have *some* idea when they need to be at each aid station
* Plan drop bags carefully with a range of possible speeds so you don’t end up in the dark, cold, or… ?
* Do at least one night trail run to debug your lights
